Semax & Selank Combo brings together two synthetic regulatory heptapeptides that have been investigated in neuroscience, neurochemistry and behavioral research. They are often discussed side by side. They are not versions of the same molecule.

Semax has the amino-acid sequence Met-Glu-His-Phe--Gly-Pro (MEHFPGP) and is also known in the scientific literature as ACTH(4–7)PGP. It was developed from a fragment of adrenocorticotropic hormone, with the ProGly-Pro sequence added at the C-terminus. Selank. Thr-Lys--Arg-Pro-Gly-Pro (TKPRPGP). Follows a different route. It is an analogue of tuftsin an endogenous tetrapeptide associated with immune regulation.

That difference matters. Research on ACTH(4-7)PGP has looked at signaling, gene expression, cerebral ischemia and neural recovery. The tuftsin-derived peptide has drawn interest in studies of anxiety-related behavior, neurotransmission and stress responses. There is some overlap. The experimental profiles are not the same.

Both molecules have a -Gly-Pro motif. Scientists have also studied their interaction with peptide-degrading enzymes; in vitro experiments, with serum showed that Semax and Selank stopped enzymes that break down enkephalin even though their measured inhibitory concentrations were different.

Why Semax & Selank Combo

The rationale for studying these compounds together comes largely from their different experimental profiles rather than from established evidence of synergy.

Semax has been investigated particularly closely in connection with neurotrophin signaling. In animal models of cerebral ischemia, researchers reported changes involving genes associated with BDNF, NGF and their receptors following administration of ACTH(4-7)PGP. Other experimental work has examined its effects on molecular pathways associated with inflammatory responses and neural injury.

The picture is still developing.

Limited human research has also been conducted with Semax as an individual compound. One study involving patients undergoing rehabilitation following ischemic stroke reported changes in plasma BDNF alongside improvement on a functional assessment scale. Those findings concern Semax alone. They do not establish an equivalent effect for a Semax-Selank combination.

Selank research has followed another path. In one animal study, researchers analyzed 84 genes associated with neurotransmission in the frontal cortex of rats. Significant expression changes were observed in dozens of genes after administration, and the investigators proposed modulation of the GABAergic system as one possible mechanism.

Cell research adds another wrinkle. Experiments using human neuroblastoma IMR-32 cells did not find the same direct gene-expression response to Selank alone. When the peptide was studied alongside GABA, however, the pattern changed substantially. The results suggest that its relationship with GABAergic signaling is more complicated than a simple direct effect on one receptor or pathway.

Human evidence for Selank is limited. A study of 62 patients with generalized anxiety disorder or neurasthenia compared the peptide with medazepam. Researchers reported similar anxiolytic effects on the assessment scales used in the study, while additional differences were observed in measures related to asthenia and enkephalin activity. The sample was small. It also investigated Selank independently, not a peptide blend.

Direct comparative research involving both molecules is thinner still. A resting-state fMRI study in 52 healthy participants examined the effects of Semax, Selank and placebo on functional connectivity involving predefined brain regions, including the amygdala and dorsolateral prefrontal cortex. Researchers found both shared and peptide-specific connectivity changes.

That makes the comparison interesting. It does not demonstrate that combining the two compounds produces stronger, additive or synergistic effects.

Research applications

Published research points to several areas in which these regulatory peptides have been investigated:

Neurotrophic signaling. ACTH(4-7)PGP has been examined for changes involving BDNF, NGF and related receptor systems, particularly in experimental models of cerebral ischemia.
Cerebral ischemia and neural recovery. Animal studies have explored molecular and transcriptional responses following ischemic injury. Limited human research has also examined Semax during post-stroke rehabilitation.
GABAergic neurotransmission. Selank studies have investigated gene-expression changes involving GABA receptors, transporters, ion channels and other components of neurotransmission.
Anxiety-related research. TKPRPGP has been studied in behavioral experiments and in limited human research involving generalized anxiety disorder and neurasthenia.
Enkephalin metabolism. In vitro experiments have examined the ability of both heptapeptides to inhibit enzymes involved in enkephalin degradation.
Functional connectivity. Human fMRI research has compared Semax and Selank in relation to resting-state connectivity involving the amygdala, dorsolateral prefrontal cortex and temporal cortical regions.

For combination research, one obvious question remains unresolved: does studying the two compounds together produce a response that differs from either peptide alone?

There is no reason to assume the answer in advance. A controlled experimental design can compare the combination with Semax alone, Selank alone and an appropriate control, allowing any differences to be measured rather than inferred from studies of the individual compounds.

Available forms

Semax & Selank research materials can be prepared in several forms depending on formulation and experimental requirements:

Lyophilized powder in a vial – a dry format suitable for storage, reconstitution and laboratory protocols that require control over solution preparation.
Reconstituted research solution – a liquid preparation for protocols in which the material needs to be available in solution.
Nasal spray formulation – relevant to experimental designs investigating intranasal delivery, a route used in published research involving these peptides.
Pre-filled syringe – a prepared liquid format that can reduce additional handling and preparation steps.
Syringe pen – a compact format with calibrated delivery settings. It can simplify repeated volume selection, reduce manual preparation steps, support more consistent measurements and limit unnecessary handling of the prepared material.

The formulation used in a study can affect stability, handling and experimental design. Results obtained with one preparation or route should not automatically be treated as equivalent to those obtained with another.

The evidence base is uneven. Mechanistic questions have been investigated in cell and animal models, while some human research exists for the individual compounds. Direct evidence for the combined formulation remains much more limited.
