What it is PT-141, or bremelanotide, is a synthetic cyclic peptide developed from Melanotan II. Its activity involves the melanocortin system, a group of receptors found throughout the body and brain. PT-141 can interact with MC1R, MC3R, MC4R and MC5R. MC4R became especially interesting to researchers because of its role in the brain. Work on PT-141 has therefore looked closely at central signalling and how it may influence sexual desire and arousal. This is quite different from studying compounds whose effects depend mainly on changes in peripheral blood flow. How it works and what the research shows There is no single receptor that explains all of PT-141's activity. Several melanocortin receptors respond to the peptide, although MC4R has drawn particular attention in research on sexual behaviour and related brain signalling. Bremelanotide has been studied not only in laboratory and animal models, but also in human clinical trials. Two large studies included more than 1,200 premenopausal women with hypoactive sexual desire disorder. Some participants received bremelanotide, while the control group received a placebo. Researchers compared changes in sexual desire andthe distress associated with low desire. Statistically significant differences between the groups were reported for both measures.
