What it is Melanotan II is a synthetic cyclic peptide based on alpha-melanocyte-stimulating hormone (α-MSH), a natural signalling peptide in the melanocortin system. It interacts with four melanocortin receptors: MC1R, MC3R, MC4R and MC5R. These receptors have different roles in the body, which is why Melanotan II has been studied in several quite different areas rather than for a single biological pathway. How it works and what the research shows MC1R is closely connected with pigmentation. When this receptor is activated in melanocytes, it stimulates melanogenesis – the process by which these cells produce melanin. This has made pigmentation one of the main areas of Melanotan II research. MC3R and MC4R are more closely associated with the central nervous system. Studies involving these receptors have looked at appetite, energy balance and sexual signalling. Animal experiments with Melanotan II have also examined changes in food intake and the wider role of melanocortin signalling in energy regulation. MC5R adds another area of interest. It is found in several tissues and has been linked to exocrine gland function, including the regulation of sebaceous glands. Melanotan II has also been tested in small human studies.
